Centrifugal microfluidic sample lysis and homogenization platform for nucleic acid extraction

Abstract


  • The application of microfluidic technologies for nucleic acid diagnostics continues to expand. While attention has primarily focused on nucleic acid amplification and detection, little consideration has been paid to sample preparation. Here, a centrifugal microfluidic platform for mechanical sample lysis and homogenization is presented. The system performs sample lysis through a magnetically-actuated beadbeating system followed by centrifugal clarification. Biological validation was performed using Bacillus subtilis spores. The platform is as efficient as in-tube beadbeating lysis and homogenization for nucleic acid extraction, and capable of processing 4 samples to near PCR-ready products in under 6 mins. © 2009 CBMS.
