Changes of superoxide dismulase activity and malondiadehyde content in brain tissue in neonatal rats with cerebral hypoxia ischemia after He-Ne laser acupuncture point irradiation

Abstract


  • Aim: To observe the changes of superoxide dismulase activity and malondiadehyde content of brain constitution after He-Ne laser acupuncture point irradiation in neonatal rats with cerebral hypoxia ischemia. Methods: The experiment was done in Department of Human Anatomy of Collage of Basic Medical Sciences of Zhengzhou University between March and November 2003. Thirty healthy Wistar rats at the age of 7 days with 12-15 g body mass, no matter male or female, were randomly divided into three groups with 10 rats in each group: sham operation control group; hypoxia ischemia group (H-I group); laser acupuncture point irradiation group. The rats in H-I group and laser acupuncture point irradiation group were made into hypoxia and ischemia model, and treated with ligation of common carotid artery under clearmindedness situation. They were put into hypoxia granary with normal pressure after resuming 1 to 4 hours under room temperature post-operation. Filling with mixture gas of 8 mL/L dephlogisticated air and 920 mL/L nitrogen fumes (volum ratio), 2 hours later the rats were took out. The rats in H-I group did not take part in any operation. The rats in laser acupuncture point irradiation group 2 days after hypoxia and ischemia were treated with Helium and Heon laser to irradiate Paihui (Du 20) and Tachui (Du 14), with 4 mW power, and 3 mm light spot diameter and 10 minutes every time and once per day at the same time and 10 days for 1 course, totally for 3 courses with every 3 days one course. Paihui was in the middle of calveria, and Tachui in the middle of back between C7-T1. The rats in sham-operated group only treated with the detachment on left-side common carotid artery without deligation, hypoxic. Rats in every group after 34 days routine breeding, the activity of superoxide dismutase (SOD) tested by xanthine oxidase and the content of malondialdehydein (MDA) was examined. Results: Because hypoxia and ischemia cause injury on animals, 3 rats in H-I group died, and 2 rats in laser acupuncture point irradiation group died. Ten, seven and eight in sham operation control group, H-I group, and laser acupuncture point irradiation group were involved in results analysis respectively. 1 The activity of SOD: It was significantly lower in H-I group than that in sham operation control group and laser acupuncture point irradiation group [(59.70±4.03) NU/mL, (76.24±2.43) NU/mL, (72.56±5.60) NU/mL,(t=5.03, 4.83, P < 0.05)]. 2 The content of MDA of brain constitution in rats: It was significantly higher in H-I group than that in sham operation control group and laser acupuncture point irradiation group [(267.58±30.55) mmol/L, (99.87±15.48) mmol/L, (135.29±32.76)mmol/L, (t=10.68,8.81, P < 0.05)]. Conclusion: He-Ne laser acupuncture point irradiation has significant positive effects on the increase of SOD activity and the decrease of MDA content in rat brain tissue after H-I. The compliance of immaturity brain in neonatal rats is huge. If the key period is applied to proceed He-Ne laser acupuncture point irradiation, which maybe have certain help on the recovery of brain function.
